what is the regulation that make us make insulation test at 500V d.c or 1000V d.c?

There are many and it depends on the country and what you are performing the test on (is it a small portable gas generator, a large 1MVA mobile diesel generator, part of another appliance or vehicle, part of a large power station, etc).

So, what country are you talking about, and what product or equipment are you intending to perform the test on?
