Reversing Aging

07/13/2023 5:59 PM

..."BUFFALO, NY- July 12, 2023 – In a groundbreaking study, researchers have unlocked a new frontier in the fight against aging and age-related diseases. The study, conducted by a team of scientists at Harvard Medical School, has published the first chemical approach to reprogram cells to a younger state. Previously, this was only achievable using a powerful gene therapy.

The team's findings build upon the discovery that the expression of specific genes, called Yamanaka factors, could convert adult cells into induced pluripotent stem cells (iPSCs). This Nobel Prize-winning discovery raised the question of whether it might be possible to reverse cellular aging without causing cells to become too young and turn cancerous.

In this new study, the researchers screened for molecules that could, in combination, reverse cellular aging and rejuvenate human cells. They developed high-throughput cell-based assays to distinguish young cells from old and senescent cells, including transcription-based aging clocks and a real-time nucleocytoplasmic protein compartmentalization (NCC) assay. In an exciting discovery, the team has identified six chemical cocktails that restore NCC and genome-wide transcript profiles to youthful states and reverse transcriptomic age in less than a week."...

Aren't most cancer tumors basically cells that refuse to age or die? Look up the Immortal Life of Henrietta Lacks .

Yes, that's my understanding as well...., apparently they are not trying to make the cells immortal which might be the difference, just reset to a younger state...So you would probably have several treatments over time...at least that's my take on this...

..."The Harvard researchers previously demonstrated that it is indeed possible to reverse cellular aging without uncontrolled cell growth by virally-introducing specific Yamanaka genes into cells. Studies on the optic nerve, brain tissue, kidney, and muscle have shown promising results, with improved vision and extended lifespan observed in mice and, recently, a report of improved vision in monkeys.

The implications of this new discovery are far-reaching, opening avenues for regenerative medicine and, potentially, whole-body rejuvenation. By developing a chemical alternative to age reversal via gene therapy, this research could revolutionize the treatment of aging, injuries and age-related diseases and offers the potential for lower costs and shorter timelines in development. On the heels of positive results in reversing blindness in monkeys in April 2023, preparations for human clinical trials of the lab’s age reversal gene therapy are in progress."...

When has any new medical treatment pioneered in the USA resulted in lower costs? Here in the USA, we seem to be proud of having the most expensive health care for the least health improvement.

Well gene therapy is very expensive with an average cost between $1 - 2 million, any cheaper alternative would be an improvement..Medical costs are high for several reasons, we live in an overly litigious society and insurance costs are through the roof....

Medical research is getting more and more expensive because of the clinical trials that must be run to get approval of the new drugs coming to market...The diagnostic expertise has steadily improved over time so that maladies that were misdiagnosed before and left untreated or even mistreated are now being properly treated...

...so these and other contributing factors like inflation and improved facilities and others is why medical insurance is so expensive...Remember those that have the money to pay for medical care must also support those that don't have the money to pay to keep the hospitals open...

MRI is much cheaper than exploratory surgery. MRI was developed in the U. S. by Dr. Raymond Damadian.

There was a company called GenRon that discovered certain chemicals that would make cells immortal, by reducing the shortening of the telemeres.

which are at the end of the e DNA strand, and after each cell replacement, it normally gets shorter. After so many regenerations it dies or turned cancerous.

There also seemed to be a "Clock" factor that also affected this, but it's source was undetermined at the time.

A liver cell, heart cell, or any other cell could become immortal, but the problem was how to stop it from growing endlessly. The company was bought out by a large pharmaceutical company, but I do not remember which one.

OOPS! Correction to post: I had not searched for Geron recently, but a recent search revealed this:

https://www.geron.com/

In the original announcement, they recommended two chemical supplements: One was ALA(Alpha Lipoic Acid) but I cannot seem to remember the other one. Anyone know or remember?

Both of these reduced the shortening of the telmeres somewhat.

I think they concentrate on creating telomere blockers as a method of killing cancer cells...as you can imagine this requires precise targeting of cancer cells only, and therein lies the problem....they currently have a drug called Imetelstat that is in various clinical trials....

https://ascopost.com/news/january-2023/phase-iii-imerge-trial-of-imetelstat-in-lower-risk-mds/#:~:text=Among%20118%20patients%20treated%20with,of%20those%20treated%20with%20placebo.

..."Imetelstat is a 13-mer oligonucleotide complimentary to the template region of the telomerase RNA component. It binds with high affinity to the template region of the RNA component of telomerase, resulting in direct, competitive inhibition of telomerase enzymatic activity.

Preclinical studies have indicated that imetelstat inhibits telomerase activity (TA) and cell proliferation of several cancer cell lines and human tumors in mouse xenograft models.10-17

Moreover, imetelstat also inhibits proliferation and induces apoptosis of cancer stem cells.18,19 Imetelstat has been evaluated in phase 1 and 1/2 clinical trials in patients with solid tumors (eg, breast and lung cancer)20,21 and hematologic malignancies (eg, chronic lymphoproliferative diseases,22 refractory and relapsed multiple myeloma,23 and myeloproliferative neoplasms [MPN]).24,25

Although in the majority of these studies imetelstat has failed to show meaningful clinical activity, phase 2 studies have demonstrated that imetelstat can elicit hematologic and molecular responses in patients with ET who are intolerant or unresponsive to prior standard therapies.24

Moreover, an additional clinical trial in MF patients revealed that imetelstat therapy could achieve complete clinical remissions in some patients using well-established clinical criteria.25

Treatment with imetelstat led to the reversal of bone marrow (BM) fibrosis and induction of morphologic and molecular remissions in some patients with MF.25 These results suggest that imetelstat has disease-modifying activity.

The precise mechanism by which imetelstat induces such responses in MPN patients has been poorly defined to date. In this report, we investigated whether imetelstat selectively targets MF HSCs and hematopoietic progenitor cells (HPCs)."....

The original lecture I saw involved taking the nucleus out of a cancer cell, and replacing it with a normal cell nucleus.

Everyone thought that the cell would become cancerous, but to their surprise, it did not It simply retained the cancer cell's tenacity for extended life span.

They tested it with various cell nucleuses with the same effect.

The telemeres retained their original length, and did not deteriorate over hundreds of regenerations.

The key seemed to be the telomere length.

The chemicals involved in preserving the telemeres were Alpha Lipoic Acid and Acetyl L Carnitine combined in specific ratios.

This was many years ago, probably more than 20 years, so evidently the research has taken another path since then.

Pharmaceutical companies do not want a one-pill-fixes-all, they want something that has to be taken regularly to maintain the benefits.

Antibiotics and vaccines are usually one time things, we have a lot of those...and surgery of course....It's a lot easier to find something that eases the symptoms of a disease than an actual cure...but there's no denying these are the three big guns...

If I recall, the other antioxidant was Acetyl-L-Carnitine (ALC) with Alpha Lipoic Acid (ALA). Now if I could only figure out the right doses...

The discovery of viagra was the beginning of reversing the aging process. (Hey hey). Now, staying young may be for everyone (that has a million or so)!
