As far as I know, 50 fold generally means 50 times (50x). It is often used in phrases like "There has been a 50 fold increase in ....".

Though it's entirely possible 50 fold means something different in the document your reading. Can you provide the sentence "50-fold" appears in? I think that would help me know for sure I'm telling you the correct definition.

Thanks. Below the sentence.

"the sample should be diluted quantitatively 50-fold with destilled water."

You Wrote: "the sample should be diluted quantitatively 50-fold with destilled water."

It definitely means 50x more water than your sample. Does it say if it's by weight or volume?

By volume - 100ml.

So I think I should dilute it in 5 liters of water. Am I right?

Well, not exactly. You should dilute to a total volume of 5 l., not just adding 100 ml to 5l.
