can grease be applied on the groove of a carbon steel ring type joint flange for installing rtj gasket?

I have used a very small ammount of silicone grease to hold the gasket in place whilst i position things so I would say yes, except if the grease would prove detrimental to the substance in the pipe. Hope this helps

If it's an oxygen line (and possibly some other fluids), no oils are greases are allowed. In most other cases, anti-seize or a lubricant compatible with the piped fluid can be used, and is often recommended.

A carbon steel ring is unacceptable for an Ultra High Vacuum chamber flange with any kind of outgassing grease. You should have learned that in vacuum 101.
