Check your OS drivers, plug and play etc. Also USB storage may be disabled.

Also, if you haven't already, try a cold start and see if the OS will recover by itself. Windows does patches all the time and maybe one of them somehow hosed the USB port. That's worked for me in the past but there are so many things that can go wrong somebody ought to write a book.

Anyway, try googling something like:

"usb drive not recognized" (1,000,000+ results) or

"usb drive not working" (5,280,000 results) or

"usb drive not being read" (4,720,000 results)

A great many of these will send you off to look at the hardware manager. Be extremely careful and read any directions before you start tinkering with the drivers and hardware settings. And, unless you're a computer genius, I'd stay away from anything to do with the registry. That's just asking for trouble.
