why do you need to consider ? a check is a check to safeguard back-pressure period.
Isolation is set of 3 valves/ 2 valves(if a check valve is there) to close and isolate to take off/do maintenance of a say, steam trap, pump, control valve, PRV etc., and temperoray operation with the by-pass.
Why complicate yourself unnecessarily. If you consider your check valve as isolation and a new guys come in he might make the mistake of your "consideration".

"Can we consider check valve as a isolation valve during operating
condition ?"
No!!!! You cannot consider a Check Valve as Isolation.
A Check valve is only capable of flow restriction in one direction. This restriction is not 100 percent. They are, by there design prone to some leakage back from the downstream side to the upstream side.
The only true "Isolation" methods for safety are:
1. Two block valves with a removable spool piece. The spool piece is between the two block valves and is fitted with a bleed valve to vent off pressure. The valves are closed, the vent is opened, when safe the spool piece is removed.
2. Double block valves with a Spec-Blind between them.
